I thought the main difference is one supports AMD cards while the other supports NVidia cards? Someone correct me if I'm wrong.

Well, both are made for gaming, and their main goal is to eliminate screen tearing.
We used to have V-Sync to do that, but it limited our FPS and negatively affected GPU performance.
Now we have G-Sync or Freesync which to that, by synchronizing your monitors refresh rate with the GPU FPS output.

The difference is that G-Sync works with Nvidia GPUs and Freesync works with AMD GPUs.

Which one is best for gaming?
Well, they both perform very similarly, but I think some tests showed that Freesync has a tiny bit less input lag, but it's negligible, especially when you consider that Nvidia GPUs perform better overall.

    Freesync is AMD. Therefore nope!

    No.

You need a Nvidia GPU and a G-Sync enabled monitor to use G-Sync.
You need an AMD GPU and a Freesync enabled monitor to use Freesync.
